In the last few years, there have been 5 notable new drugs approved for prostate cancer, including one drug combination. Let's have a look at each of them.

#1: Xtandi (enzalutamide)

Xtandi has been approved for use in late-stage castration-resistant prostate cancer since 2012 1. Since then, it has gained additional approvals as well, as follows:

  • for treating non-metastatic castration-resistant prostate cancer (CRPC);
  • for treating metastatic castration-sensitive prostate cancer (mCSPC);
  • for treating non-metastatic castration-sensitive prostate cancer (nmCSPC) with biochemical recurrence (BCR) at high risk of metastasis. Xtandi is the first androgen receptor signaling inhibitor approved in this earlier stage of prostate cancer. It can be combined with a gonadotropin-releasing hormone (GnRH) analog or used as a standalone treatment 2.

Is Xtandi available in Nigeria?

Xtandi (enzalutamide) is not registered in the NAFDAC database. However, some online pharmacies in Nigeria seem to offer the medicine. #2: Orgovyx (relugolix)

A newer medicine than Xtandi, Orgovyx was first approved in 2020 for use in advanced prostate cancer 4. Orgovyx is an oral medicine, easy to take at home, with or without food.

How successful is Orgovyx for prostate cancer?

According to the results of its clinical trial:

  • 97% of patients taking Orgovyx lowered their testosterone within 29 days and kept it low until week 48 of treatment;
  • After 15 days of treatment, 99% of men taking Orgovyx lowered their testosterone to levels under 50 ng/dL. Only 12% of the patients in the control group reported the same result.
  • Prostate-specific antigen levels (PSA) after 3 months of treatment were 92% lower, on average, for patients taking Orgovyx vs the control group 4.

Is Orgovyx available in Nigeria?

Orgovyx isn't registered in the NAFDAC database. However, this doesn't mean that you cannot access it. #3: Lynparza (olaparib)

  • Olaparib for metastatic castration-resistant prostate cancer with a BRCA mutation

    • The risk of cancer progression was 76% lower for patients taking Lynparza with abiraterone and prednisone 6.
  • Olaparib for metastatic castration-resistant prostate cancer with an HRR mutation, as well as BRCA1, BRCA2, or ATM mutation

    • The risk of cancer progression was 66% lower for patients taking Olaparib vs the control group;
    • 33% of patients taking Lynparza saw their tumors shrink, compared to 2% in the control group 6.

Is Lynparza available in Nigeria?

#4: Akeega (niraparib/abiraterone acetate)

How successful is Akeega for prostate cancer?

  • The median overall survival for patients taking Akeega was 30.4 months, compared to 28.6 months in the control group 8.

Is Akeega (niraparib/abiraterone acetate) available in Nigeria?

#5: Xtandi (enzalutamide) + Talzenna (talazoparib)

As of 2023, Xtandi is also approved in combination with Talzenna (talazoparib) for use in patients with HRR gene-mutated metastatic castration-resistant prostate cancer (mCRPC) 9.

How successful is enzalutamide plus talazoparib for prostate cancer?

According to the recently reported results of the TALAPRO-2 study, the combination of these two medicines significantly improves survival in patients, compared to Xtandi (enzalutamide) alone 10. Additional key findings were:

  • 67% of patients treated with Xtandi + Talzenna had their tumors shrink or disappear, compared to 40% in the control group;
  • Patients with BRCA1/BRCA2 mutations had an 80% lower risk of cancer progression following treatment with Xtandi + Talzenna. For patients without the gene mutation, the risk was lowered by 32% 10.

Is Talzenna (talazoparib) available in Nigeria?
